Testing of ornamental and crop plants for infestation with pospiviroids

RLP AgroScience has successfully participated in the FAPAS Proficiency Test PL0102 (Laboratory 002) and has been recognised by the Federal Biological Research Centre as a laboratory for viroid testing.
Potato spindle tuber viroid (PSTVd) has been repeatedly and sporadically detected in potato and tomato crops in various EU member states in recent years. In most cases, the cause of the occurrence could not be clarified. In spring 2006, PSTVd was detected for the first time in the Netherlands as part of a monitoring programme on ornamental plants of the Solanaceae family, initially on Solanum jasminoides and later also on Brugmansia spp. In no case were symptoms visible on the infested plants. Based on further test results in summer/autumn 2006 in the Netherlands and other EU member states (BE, DE, FR, SI), to which deliveries were made from Dutch infested farms, there is considerable reason to suspect that PSTVd could also be widespread in ornamental plant species in Germany. Initial tests from our institute show that not only infections with PSTVd occur but also with the Tomato apical stunt viroid (TASVd) (Verhoeven et al., 2008).
Studies on Potato spindle tuber viroid (PSTVd), Tomato chlorotic dwarf viroid (TCDVd), Tomato apical stunt viroid (TASVd), Chrysanthemum stunt viroid (CSVd), Citrus exocortis viroid (CEVd), Columnea latent viroid (CLVd), Mexican papita viroid (MPVd), Tomato planta macho viroid (TPMVd), Irisine viroid 1 (IrVd-1) and Pepper chat fruit viroid (PCFVd) are routinely carried out in our institute using RT-PCR. Testing for infestation with other viroids is possible on request:

1) The basic price refers to the detection of a specific viroid (1-2 primer pairs), for a test for general viroid infestation (2-4 primer pairs), €84.00 is charged instead of €42.00.
2) The surcharge for individual samples and the individual report does not apply from a number of samples >5 (reason: the same controls must be carried out for individual samples and a complete report must be prepared).
3) Reason for additional costs: if mixed samples are sent in, a part must be taken from each cutting or leaf and combined with the others (additional manual labour). If mixed samples are to be compiled, please ensure that only cuttings or only one leaf is taken from each plant. We only take part of a leaf from a cutting and combine 25 leaf pieces into one sample. Leaves that have fallen off during transport are not taken into account. If leaves are submitted, we also take only a part of each leaf and again combine 25 pieces into one sample. If several leaves were harvested from one plant, the number of 25 samples would increase.
4) Reason for the additional costs: there are still no standardised regulations for the testing of seeds. The extraction of RNA from seeds often causes problems. In order to ensure a reliable diagnosis for the samples, we allow the seeds to germinate and then analyse samples of the first primary leaves. Plant cultivation requires additional labour (sowing, harvesting of samples), resources (greenhouse) and material (soil, pots). There is therefore an additional charge of €18.00 per seed sample.
